Generation of recombinant rotaviruses encoding a split NanoLuc peptide tag

Recombinant viruses expressing fluorescent or luminescent reporter proteins are used to quantitate and visualize viral replication transmission. Here, we a split NanoLuc luciferase (NLuc) system comprising large LgBiT small HiBiT peptide fragments generate stable rotaviruses (RVs). Reporter RVs NSP1-HiBiT fusion protein were generated by placing an 11 amino acid tag at the C-terminus of intact simian RV NSP1 open reading frame truncated human frame. Virus-infected cell lysates exhibited NLuc activity that paralleled virus replication. The antiviral neutralizing antibodies reagents against recombinant monitored measuring reductions in expression. These findings demonstrate systems powerful tools for studying life cycle pathogenesis, robust platform developing novel drugs.
